Abstract

Pod pojmom restrikcijska analiza, u molekularnoj genetici i genetičkom inženjerstvu, podrazumijeva se analiza DNA korištenjem restrikcijskih enzima, odnosno endonukleaza koje cijepaju DNA u točno određenim sekvencijama. Stoga su ovi enzimi nezamjenjivi u genetičkom inženjerstvu i mnogim molekularno-genetičkim metodama koje imaju primjenu u dijagnostici, filogeniji i identifikaciji organizama. U ovom radu je restrikcijskom analizom potvrđena struktura prethodno konstruiranih plazmida pRS40cN, pRS52cN, pRS53cN i pRS55cN. U tu su svrhu plazmidi najprije izolirani i pročišćeni iz velikih volumena kultura bakterije Eschericha coli, a zatim pocijepani odgovarajućim restrikcijskim enzimima te podvrgnuti elektroforezi u agaroznom gelu. Na temelju rezultata analize potvrđeno je da restrikcijske mape svih plazmida odgovaraju njihovoj stvarnoj strukturi.

Abstract (english)

In molecular genetics and genetic engineering, the term restriction analysis refers to the analysis of DNA molecules using restriction enzymes, endonucleases that cleave DNA in specific nucleotide sequences. Therefore, these enzymes are indispensable in genetic engineering, as well as in many molecular genetic methods that are used in diagnostics, phylogeny and identification of organisms. In this paper, restriction analysis was used to confirm the structure of previously constructed plasmids pRS40cN, pRS52cN, pRS53cN and pRS55cN. For this purpose, the plasmids were first isolated and purified from large volumes of Escherichia coli cultures, then digested with appropriate restriction enzymes and subjected to agarose gel electrophoresis. Based on the results of the analysis, it was confirmed that the restriction maps of all plasmids correspond to their actual structure.
