Abstract

Kvasac Saccharomyces cerevisiae je modelni organizam u biokemijskim i genetičkim istraživanjima zbog sličnosti mnogih staničnih procesa sa procesima u višim eukariotima. Stanice kvasca obavija stanična stijenka građena od glukanske mreže, koju čine β-1,3 i β-1,6- glukan, hitin i manoproteini. Građa stanične stijenke mijenja se ovisno o staničnom ciklusu kvasca pa se tako razlikuje u mitozi i mejozi. Prelaskom iz mitoze u mejozu, a zatim i prilikom formiranja sporula, dolazi do značajnih promjena u strukturi stanične stijenke što je posljedica promijenjene ekspresije pojedinih proteina. Cilj ovog rada bio je obilježiti proteine stanične stijenke Pir1, Pir3, Pir5 i Spr1 hemaglutininskom oznakom kako bi olakšali njihovo praćenje u različitim fazama staničnog ciklusa.

Abstract (english)

Yeast Saccharomyces cerevisiae is a model organism in biochemical and genetic research due to the similarity of many cellular processes with those in higher eukaryotes. Yeast cells are surrounded by a cell wall built of a glucan network, made up of β-1,3 and β-1,6-glucan, chitin, and mannoproteins. The structure of the cell wall varies depending on the cell cycle of the yeast, so it differs in mitosis and meiosis. With the transition from mitosis to meiosis, and then spore formation, there are significant changes in cell wall structure as a result of altered expression of individual proteins. The aim of this study was to label cell wall proteins Pir1, Pir3, Pir5 and Spr1 with a hemagglutinin label to facilitate their monitoring at different stages of the cell cycle.
