Abstract

Alge su organizmi koji mogu u najvećoj mjeri apsorbirati i skladištiti teške metale. U ovom radu je proučavana toksičnost soli teških metala na algu Euglena gracilis, koja je poslužila kao biotestni mikroorganizam. Kinetika nakupljanja prolina i stvaranje cista (palmela stadij) u E. gracilis proučavane su tijekom 12 dana uzgoja pri različitim koncentracijama soli teških metala (Cu2+, Pb2+, Co2+ and Hg2+ ;0.5 do 2.0 mM). Izloženost E. gracilis povećanim kocentracijama teških metala dovela je do njihovog nakupljanja u stanicama. Posljedica toga bilo je nakupljanje prolina, kao i pojava palmela stadija u staničnom ciklusu E. gracilis, ovisno o koncentraciji teških metala u životnom okolišu. Uočeno je da što je veća toksičnost ili nakupljanje metala, to je veći udjel intracelularnog prolina u stanicama alga. Nakupljanje prolina započelo je nekoliko sati nakon izlaganja metalima, dok je stvaranje cista uočeno nakon nekoliko dana izloženosti.

Abstract (english)

Algae are the main organisms that take up and store heavy metals. The present study was aimed at the toxicity evaluation of heavy metals, using the flagellate Euglena gracilis as a biotest organism. Parameters of the flagellate like proline accumulation and palmella stage were used as end points for the toxicity assessment. E. gracilis was exposed for 12 days to Cu2+, Pb2+, Co2+ and Hg2+ from 0.5 to 2.0 mM. Exposure of E. gracilis to elevated concentration of heavy metals led to intracellular accumulation of high concentrations of these metals in cells. Concomitantly, accumulation of free proline occurred, as well as palmella stage od E. gracilis life circle, depending on the concentration of heavy metals in the external medium or in the cell. The greater the toxicity or accumulation of a metal, the greater the amount of intracellular proline in algal cells.The accumulation of proline was triggered within a few hours of metal treatment while the cysts formation occurs after few days of treatment.
