Abstract

Proizvodne stanične linije važan su ekspresijski sustav za proizvodnju biofarmaceutika koji se primijenjuju u terapiji mnogih oboljenja poput autoimunih bolesti, hematoloških poremećaja, hormonalne neravnoteže, karcinoma, infekcija, genetičkih poremećaja i dr. Kako bi se zadovoljili rastući zahtjevi za biofarmaceuticima te povećala ekonomska isplativost proizvodnje važno je optimirati proces proizvodnje obzirom da se kulturom životinjskih stanica dobivaju niži prinosi proizvoda u odnosu na druge ekspresijske sustave. Neke od strategija koje se primjenjuju u tu svrhu su kontrola staničnog ciklusa, stanične smrti i metabolizma stanica zbog čega je vrlo važno poznavati mehanizme tih staničnih procesa. Navedene strategije pokazale su veliki potencijal primjene u proizvodnji biofarmaceutika. U ovom radu dan je pregled najčešće korištenih proizvodnih staničnih linija, procesa stanične diobe i smrti te kako se kontrolom navedenih staničnih procesa mogu ostvariti poboljšanja u proizvodnji biofarmaceutika.

Abstract (english)

Production cell lines are an importatnt expression system for the production of biopharmaceuticals that are used in the treatment of many diseases such as autoimmune diseases, hematological disorders, hormonal imbalance, cancer, infections, genetic disorders etc. In order to meet growing demands for biopharmaceuticals and increase the economic profitability of production, it is important to optimize the production process, since animal cell cultures result with lower production yields compared to the other expression systems. Some of the strategies applied for this purpose are control of cell cycle, cell death, and cell metabolism, so it is very important to know the mechanisms of these cellular processes. These strategies have shown great potential for application in the production of biopharmaceuticals. This graduation thesis provides an overview of the most commonly used production cell lines, cell division and death and how improvements in the production of biopharmaceuticals can be achieved by controlling these cellular processes.
