Abstract

Escherichia coli jedna je od najpoznatijih i najčešće korištenih mikroorganizama za proizvodnju rekombinantnih terapijskih proteina zahvaljujući temeljitom poznavanju genoma bakterije, velikoj brzini rasta i gustoći stanica, laganoj prilagodljivosti mikroorganizma procesima industrijskih razmjera i sveopćoj ekonomičnosti procesa. U ovom radu opisan je proces proizvodnje rekombinantnih terapijskih proteina, od ekspresije ciljanog proteina u samoj bakteriji, tehnika razbijanja stanica, izolacije i otapanja inkluzijskih tijela do renaturacije proteina i ponovnog smatanja u nativnu konformaciju. Nadalje, na primjeru proizvodnje rekombinantnog terapijskog proteina FGF21 opisane su mogućnosti poboljšanja procesa s ciljem optimizacije prinosa proizvoda uz zadržavanje zadovoljavajuće razine kvalitete ciljanog produkta.

Abstract (english)

Escherichia coli is one of the most famous and most commonly used microorganisms for the production of recombinant therapeutic proteins due to the thorough understanding of its genome, rapid growth rate and high cell density, its scalability and overall process economicity. This paper provides an insight into the process of production of recombinant therapeutic proteins from protein expression in E. coli through methods of cell disruption, isolation, solubilization of inclusion bodies, renaturation and refolding of proteins into their native conformation. Furthermore, methods of process intensification are explained using FGF21 recombinant protein production process as an example.
