Abstract

Razmatrajući nedostatke prirodnih peptida koji se mogu prevladati konformacijskim modifikacijama, novija istraživanja se fokusiraju na male molekule koje ugradnjom u peptidne lance induciraju tvorbu različitih okreta i elemenata sekundarne strukture te omogućuju pravilno nabiranje proteinskih lanaca. U sklopu ovog završnog rada pripravit će se i spektroskopski analizirati (IR-, NMR- i CD-spektroskopija) ferocenski dipeptidi Boc-D-Phe-NH-Fn-COOMe (1) i Ac-D-Phe-NH-Fn-COOMe (2) izvedeni iz ferocenske aminokiseline i fenilalanina. Dobiveni dipeptidi su pogodan model za istraživanje i nadogradnju spoznaja o utjecaju strukture i svojstava konstituirajućih aminokiselina na sekundarnu strukturu pripadajućih peptida kao i na njihov potencijalni terapeutski učinak.

Abstract (english)

Considering the limitation of natural peptides that can be overcome by conformational modifications, recent studies explore small molecules that (when incorporated into peptide chains) induce the formation of various turns and elements of secondary structure and allow proper folding of protein chains. As part of this Thesis, ferrocene dipeptides Boc-D-Phe-NH-Fn-COOMe (1) and Ac-D-Phe-NH-Fn-COOMe (2) were synthesized from the ferrocene amino acid and the phenylalanine and spectroscopically analyzed using IR-, NMR- and CD-spectroscopy. The obtained dipeptides are a useful model for new researches and upgrade of knowledge about the influence of amino acid backbones on the secondary structure of peptides as well as their potential therapeutic effect.
