Procjena toksičnosti niskotemperaturnih eutektičkih otapala za razvoj formulacije ciprofloksacina

Abstract

Eutektička otapala zelena su otapala sastavljena od donora i akceptora vodikove veze koja u određenom molarnom omjeru imaju temperaturu tališta nižu od pojedinačnih komponenti pa su najčešće pri sobnoj temperaturi u tekućem agregacijskom stanju. Komponente od kojih se pripremaju eutektička otapala nisu toksične, no potrebno je ispitati njihov sinergijski učinak kako bi se procijenio njihov utjecaj na ljude i okoliš. U ovom radu ispitana je citotoksičnost triju različitih eutektičkih otapala (limunska kiselina: glukoza, betain: jabučna kiselina: glukoza, glicerol: glukoza) kao i terapeutskog eutektičkog otapala (limunska kiselina: glukoza: ciprofloksacin) na Caco-2 i HeLa staničnu liniju MTS metodom. Otapala nisu pokazala inhibitoran učinak na rast stanica pa se mogu smatrati netoksičnim otapalima.

Abstract (english)

Deep eutectic solvents are green solvents, formed by a donor and an acceptor of hydrogen bond. At room temperatures they are usually liquids, due to the fact that in certain molar ratios their melting point is lower than a melting points of each forming compound. DESs forming compounds are considered to be non-toxic, but before their application it is necessary to determine synergic effect of forming compounds and to assess their impact on human and the environment. In this thesis, the impact of three DESs (citric acid:glucose, betaine:malic acid:glucose, glycerol:glucose) and one THEDES (citric acid: glucose: ciprofloxacin) on Caco-2 and HeLa cell line was examined using MTS method. Solvents did not show inhibitory effect on cell growth and can be considered noncytotoxic.
