Abstract

Posljednjih godina nastoji se smanjiti upotreba organskih hlapljivih otapala zbog negativnog utjecaja na okoliš. Takva otapala se pristupom zelene kemije nastoje zamijeniti prirodnom alternativom koja ne škodi ljudskom zdravlju i okolišu. Nova generacija takvih otapala su niskotemperaturna eutektička otapala (engl. Deep Eutectic solvents, DES) koja karakteriziraju jednostavna sinteza, niska hlapljivost, biorazgradivost. U ovom radu provedena je reakcija enantioselektivne redukcije halogeniranog acetofenona katalizirane enzimima pekarskog kvasca Saccharomyces cerevisiae u niskotemperaturnom eutektičkom otapalu kolin klorid:etilen-glikol s različitim masenim udjelima vode. Također, ispitana je i vijabilnost stanica te utjecaj predtretmana stanica kvasca kuglicama na konverziju reakcije i stereoselektivnost biokatalizatora. S obzirom na rezultate konverzije i enantiomerni višak DES su se pokazala kao povoljan izbor za bioredukciju kao i korištenje kvasca kao biokatalizatora u odnosu na pufer.

Abstract (english)

In recent years, efforts have been made to reduce the use of organic volatile solvents due to the negative impact on the environment. With the approach of green chemistry, such solvents are being replaced by a natural alternative that does not harm human health and the environment. A new generation of such solvents are low temperature deep eutectic solvents characterized by simple synthesis, low volatility, biodegradability. In this study, the enantioselective reduction of halogenated acetophenone catalyzed by enzymes of the baker's yeast Saccharomyces cerevisiae in low temperature deep eutectic solvents choline cloride:ethylene-glycol with different water contents was performed. Also, cell viability and the influence of yeast cell pretreatment with beads on the reaction yield and stereoselectivity of biocatalysts were examined. Given the results of utilization and enantiomeric excess DES have been proved to be a favorable choice for bioreduction as well as the use of yeast as a biocatalyst compared to buffer.
