Abstract

Ortodontske naprave primjenjuju se već niz godina kao primarni oblik postizanja ortodontskog pomaka i estetski ljepšeg pozicioniranja zubi. Najčešće su izrađene od metala, koji u usnoj šupljini lako korodiraju i ispuštaju ione u slinu. Metalni ioni imaju poznat čitav niz negativnih bioloških aktivnosti te se postavlja pitanje štete li metalni ioni ispušteni od strane ortodontskih naprava pacijentima. Cilj ovog rada bio je ispitati citotoksičan, genotoksičan i prooksidativni efekt metala ispuštenih u slinu iz dijelova ortodontskih naprava. Za mjerenje citotoksičnosti korištena je Neutral Red metoda, za mjerenje genotoksičnosti komet test, a za mjerenje prooksidativnog učinka metode DCFH-DA i linearizacije plazmida φX-174 RF I. Vrijeme eluacije ortodontskih naprava utječe na sastav sline te se mogu izmjeriti različiti biološki efekti na stanicama. Većina eluata djelovala je toksično u cijelom koncentracijskom rasponu neovisno o vremenu eluacije na CAL 27 stanice. Na AGS staničnoj liniji određeno je da niže koncentracije eluata bravica, ligatura, prstenova te svih dijelova izazivaju prooksidacijsko djelovanje, dok više koncentracije eluata istih ortodontskih naprava imaju antioksidacijsko djelovanje. Prooksidacijski učinak određen na plazmidu φX-174 RF I je bio slab. Metali prisutni u eluatima ne izazivaju trajne promjene na genetički materijal stanica te je njihova primjena sigurna.

Abstract (english)

Orthodontic appliances have been used for decades as a primary way of treating orthodontic problems and achieving an aesthetically pleasing look. They are usually made of metal, which can easily corrode and release metal ions into saliva. Metal ions are known for their numerous negative biological effects, so a question, whether ions released by appliances cause damage to patients, rises. The goal of this paper was to test cytotoxic, genotoxic and prooxidative effects of metal ions released by appliance parts into saliva. The results show that a certain amount of those effects on cells is visible, but that it is not worrying to the point of saying that orthodontic appliances are harmful for human health. Cytotoxicity was measured using NR method, prooxidative effects were tasted using the DCFH-DA method and φX-174 RF I plasmid linearization, while genotoxicity was measured with comet assay. The eluation time has an impact on the composition of the saliva and biological effects can be measured. Most of the tested samples had a toxic effect on CAL 27 cell line. Regarding AGS cell line, lower concentrations of samples were shown to have a prooxidative effect, while higher concentrations had an antioxidative effect. The prooxidative effect tested using the φX-174 RF I plasmid linearization was weak. Metals present in the eluates do not cause permanent changes of the genetic material and their application is safe.
