Abstract

Probiotici su živi mikroorganizmi, koji primijenjeni u adekvatnoj količini imaju povoljan učinak na zdravlje domaćina. Prilikom odabira probiotičkih sojeva, jedan od najvažnijih kriterija je svojstvo pojedine bakterije da adhezira i kolonizira gastrointestinalni trakt domaćina. Jedan od ciljeva ovog rada je bio ispitati utječe li prisutnost S-sloja na svojstvo adhezije Lactobacillus brevis ZG1 i Lactobacillus brevis SF15B na Caco-2 epitelne stanice, uz sojeve Lactobacillus helveticus M92 za koji je poznato da ima S-sloj (pozitivna kontrola) i Lactobacillus plantarum D13 za koji je poznato da nema površinski S-sloj (negativna kontrola). Rezultati su pokazali da je prisutnost S-sloja veoma bitan faktor za adheziju na crijevni epitel. Uz adheziju, jedan od zahtjeva za odabir probiotičkih sojeva jest i antimikrobna aktivnost među čije mehanizme ubrajamo i kompetitivnu ekskluziju prema patogenima. Ispitani su spomenuti probiotički sojevi u uvjetima predinkubacije i koinkubacije pri čemu se pokazalo da značajno smanjuju sposobnost adhezije patogena Escherichia coli i Salmonella enterica serovar Typhimurium.

Abstract (english)

Probiotics are live microorganisms which, when administered in adequate amounts, confer a health benefit on the host. In the selection of probiotics, one of the most important criteria is the ability of bacterial adhesion and colonization of the hosts gastrointestinal tract. One of the goals of this study was to determine whether the presence of S-layer has the influence on Lactobacillus brevis ZG1 and Lactobacillus brevis SF15B adhesion on Caco-2 epithelial cells, while Lactobacillus helveticus M92 which has the S-layer was the positive control and Lactobacillus plantarum D13, without S-layer, was the negative control. The results showed that the presence of S-layer is an important factor for the adhesion to the intestinal epithelium. Another requirement for the selection of probiotic strains is the antimicrobial activity among whose mechanisms is also the competitive exclusion to pathogens. Mentioned probiotic strains examined in terms of co-incubation and pre-incubation significantly reduced adhesion of pathogenic Escherichia coli and Salmonella enterica serovar Typhimurium.
