Abstract

Ljekovite biljke sadrže više različitih kemijskih tvari koje djeluju povoljno na zdravlje, pojedinačno, kumulativno ili u sinergiji s drugim tvarima. U ovom radu je ispitan kemijski i bioaktivni profil cvijeta maslačka, određen je biološki učinak ekstrakta cvijeta maslačka na Cal27, Cal27-2B1 i T24 stanice te je istražen citotoksični učinak na različite vrste laktobacila. Određen je udio ukupnih polifenolnih spojeva i pojedinih skupina te pigmentnih spojeva i topljivih polisaharida pri čemu su hidroksicimetne kiseline i karotenoidi (β-karoten) zastupljeni u visokom udjelu. Utvrđeno je da ekstrakt cvijeta maslačka ima približno isti antioksidacijski kapacitet bez obzira na primijenjenu metodu određivanja. Ekstrakt cvijeta maslačka pokazuje citotoksični učinak kod staničnih linija raka jezika, pri čemu je epitel jezika s eksprimiranim integrinom najosjetljiviji. Ekstrakt maslačka djeluje slabo prooksidativno, a indukcija slobodnih radikala ne utječe na promjenu preživljenja staničnih linija. Ekstrakt cvijeta maslačka nije toksičan za odabrane sojeve laktobacila te njegovom upotrebom neće doći do narušavanja ravnoteže mikroflore u sisavaca.

Abstract (english)

Medicinal plants contain different chemical compounds that may act individually, additively, or in synergy to improve health. In this study the chemical and bioactive profile of the dandelion flower extract was evaluated as well as its biological effect on the Cal27, Cal27-2B1 and T24 cell lines. The plant extract was evaluated for its total phenolic content, specific phenolic compounds, pigments and soluble polysaccharides. It contains a high level of hydroxycinnamic acids and carotenoids (β-carotene). The dandelion flower extract had similar antioxidant capacities despite the three different methods used. The extract exhibited citotoxic activity against the tongue carcinoma cell lines, wherein the tongue epithelium with expressed integrin was the most sensitive one. The extract showed low prooxidative effect and the free radical induction had no influence on cell viability. The antimicrobial activity against selected lactic acid bacteria was observed wherein the extract exhibited no inhibitory effect which means that the consumption of dandelion flower extract will not compromise the microbial balance in mammals.
