Abstract

Velika količina lignoceluloznog otpada, kojeg čine uglavnom celuloza, hemiceluloza i lignin, nastaju u šumarstvu, poljoprivredi i prehrambenoj industriji. Lignoceluloza je jeftina, ali i kompleksna obnovljiva sirovina velike biotehnološke vrijednosti te se može prevesti u razne visokovrijedne biotehnološke proizvode. Da bi se iskoristio potencijal lignoceluloze potrebno je primijeniti prikladne metode predobrade. Jedan od mogućih načina konverzije biomase u fermentabilne šećere za kasniju fermentaciju je metode enzimska hidroliza. Za potpunu enzimsku hidrolizu lignoceluloze potrebno je sinergijsko djelovanje raznolikih enzima. Istraživanjem interakcija mnoštva različitih enzima traže se optimalne kombinacije i omjeri enzima za određene lignocelulozne supstrate. U ovome završnome radu opisani su enzimi koji se koriste za razgranju različitih komponenata lignoceluloze. Mnogo čimbenika poput omjera enzima, koncentracija supstrata i enzima, inhibitori, adsorpcija i surfaktanti/tenzidi utjece na enzime i na optimizaciju hidrolitickog procesa.

Abstract (english)

Huge amounts of lignocellulosic waste, consisting mainly of cellulose, hemicellulose and lignin, are generated through forestry, agriculture and food industry. Lignocellulose is a cheap but complex renewable raw material of enormous biotechnological value and can potentially be converted into various value added biotechnological products. In order to exploit its potential it is necessary to implement appropriate pretreatment methods. Enzymatic hydrolysis is one of the possible technologies for the conversion of biomass into fermentable sugars for subsequent fermentation. Lignocellulose requires a variety of enzymes, acting in synergy, for its complete hydrolysis. These synergistic interactions between different enzymes have been investigated in order to design optimal combinations and ratios of enzymes for different lignocellulosic substrates. This final work describes the enzymes required to degrade various components of lignocellulose. Many factors affect the enzymes and the optimisation of the hydrolysis process, such as enzyme ratios, substrate loadings, enzyme loadings, inhibitors, adsorption and surfactants.
