Proizvodnja bioetanola i 2,3-butandiola pomoću kvasca Kluyveromyces marxianus na enzimskom hidrolizatu predobrađenog drveta bukve

Abstract

Lignocelulozne sirovine su biomasa drvenastih i nedrvenastih biljaka. Otpadne lignocelulozne sirovine imaju izniman potencijal u biotehnološkoj industriji pri proizvodnji bioetanola te drugih biokemikalija. Unatoč niskoj cijeni, lakoj dostupnosti te širokoj rasprostranjenosti, uporaba lignoceluloznog otpada kao sirovine je rijetka zbog složene strukture, otpornosti na enzimsku hidrolizu i nedovoljno razvijenih metoda predobrade. U ovom radu provedena je enzimska hidroliza (50 °C, pH 5) kiselinski predobrađenog čvrstog dijela bukve u 0,01 M acetatnom puferu, pomoću enzima Viscozyme L (5 % vol volˉ¹) i Cellulase enzyme blend (2 % vol volˉ¹). Na dobivenom enzimskom hidrolizatu proveden je uzgoj kvasca Kluyveromyces marxianus NBRC 1777 uz različite uvjete sastava podloge, pH vrijednosti (3,7; 4,4 i 5,1), stupnja aeracije te temperature (25, 29 i 33°C). Među dobivenim produktima zabilježena je određena koncentracija etanola u rasponu vrijednosti od 29,10 do 33,07 g Lˉ¹, te koncentracija 2,3-butandiola u rasponu vrijednosti od 0,52 do 0,69 g Lˉ¹.

Abstract (english)

Lignocellulosic raw materials are biomass of wood and non-wood plants. Waste lignocellulosic raw materials have exceptional potential in the biotechnology in the production of bioethanol and other biochemicals. Despite the low cost, easy availability and wide distribution, the use of lignocellulosic waste as a raw material is rare due to its complex structure, resistance to enzymatic hydrolysis and insufficiently developed pretreatment methods. In this work, enzymatic hydrolysis (50 °C, pH 5) of solid phase obtained by acid pretreatment of beech wood chips, in 0.01 M acetate buffer, was preformed using the enzymes Viscozyme L (5 % vol volˉ¹) and Cellulase enzyme blend (2 % vol vol ˉ¹). The resulting enzyme hydrolyzate was used for cultivation of yeast Kluyveromyces marxianus NBRC 1777 under different conditions of media composition, pH conditions (3.7; 4.4 and 5.1), various aeration rates and temperature conditions (25, 29 and 33 °C). Among the products obtained, a certain concentration of ethanol in the range from 29.10 to 33.07 g Lˉ¹, and a concentration of 2,3-butanediol in the range from 0.52 to 0.69 g Lˉ¹ were recorded.
