Abstract

DNA je kontinuirano izložena agensima koji uzrokuju oštećenja, a integritet DNA posebno narušavaju unakrsne veze DNA i proteina (engl. DNA-protein crosslink, DPC). Navedeno oštećenje ugrožava osnovne transakcije DNA molekule, zbog čega se kod ljudi akumulacija DPC-ova povezuje sa starenjem, nastankom karcinoma i neurodegeneracijom. Proteini Acrc, Zatt i Xpa imaju ulogu u različitim mehanizmima popravka DPC-ova. Acrc vjerojatno provodi proteolizu oštećenja, Zatt vjerojatno sudjeluje u procesu hidrolize unakrsne veze, a Xpa je dio puta popravka koji je ovisan o nukleaznoj aktivnosti. U ovome radu, provedena je ekspresijska analiza gena ACRC, ZATT i XPA u tkivima domaćeg miša (Mus musculus), te usporedba ekspresijskih profila ovisno o dobi i spolu. Također, ekspresijski profili proučavanih gena uspoređeni su sa ekspresijom kod čovjeka.

Abstract (english)

DNA is constantly exposed to agents that cause damage. DNA integrity is particularly affected by DNA-protein crosslink (DPC). DPC lesions threaten basic DNA transactions, which is why in humans the accumulation of DPCs is associated with aging, cancer and neurodegeneration. The proteins Acrc, Zatt, and Xpa play roles in various mechanisms of DPC repair. Acrc likely performs DPC proteolysis, Zatt is probably involved in the process of crosslink hydrolysis, and Xpa is part of a repair pathway that is dependent on nuclease activity. In this work, expression analysis of the genes ACRC, ZATT, and XPA was performed in the tissues of the house mouse (Mus musculus) and expression profiles were compared as a function of age and sex. In addition, the expression profiles of the studied genes were compared with the expression in humans.
