Abstract

Helicobacter pylori uzročnik je jedne od najčešćih bakterijskih infekcija u ljudi, a njezino otkriće prije 20 godina promijenilo je dijagnozu i liječenje gastroduodentitisa. Sposobnost preživljavanja u želucu povezana je višestrukim mehanizmima, a novija istraživanja upućuju na to da je formiranje biofilma povezano s kolonizacijom. Cilj ovoga rada bio je kvantificirati sposobnost formiranja biofilmova i stupanj autoagregacije H. pylori te karakterizirati izolate bakterija mliječne kiseline iz ovčjeg sira s ciljem pronalaska potencijanih probiotika. Određena je i sposobnost inhibicije biofilmova H. pylori te stupanj koagregacije izolata s ovom patogenom bakterijom u simuliranim uvjetima želuca. U eksperimentalnim uvjetima, H. pylori klasificirana je kao jak producent biofilma sa značajnim autoagregacijskim svojstvima (45%). Tri izolata pokazala su najveći stupanj inhibicije biofilma i koagregacije s H. pylori te su identificirani pomoću fermentacijskog profila kao Latilactobacillus curvatus DK1, Lacticaseibacillus paracasei DK5 i Latilactobacillus curvatus DK7.

Abstract (english)

Helicobacter pylori is the cause of one of the most common bacterial infections in humans, and its discovery 20 years ago has transformed the diagnosis and treatment of gastroduodenitis. The ability to survive in the stomach is related to several mechanisms, and recent research suggests that biofilm formation is related to colonization. The aim of this work was to quantify the ability to form biofilms and the degree of autoaggregation of H. pylori and to characterize isolates of lactic acid bacteria from sheep cheese to find potential probiotics. In addition, the ability to inhibit H. pylori biofilms and the degree of coaggregation of isolates with this pathogenic bacterium were determined under simulated gastric conditions. Under experimental conditions, H. pylori is classified as a strong biofilm producer with significant autoaggregation properties (45%). Three isolates showed the highest degree of biofilm inhibition and coaggregation with H. pylori and were identified as Latilactobacillus curvatus DK1, Lacticaseibacillus paracasei DK5 and Latilactobacillus curvatus DK7 based on the fermentation profile.
