Abstract

Cilj ovog rada bio je transformirati bakteriju Escherichia coli plazmidima pUC-GW-RadH i pUC-GW-NoxV, te potvrditi uspješnost transformacije restrikcijskom analizom plazmidne DNA izolirane iz odabranih transformanata. Navedeni plazmidi nose gene koji kodiraju za R-specifičnu alkohol-dehidrogenazu i NADH-oksidazu. Bakterija E. coli je transformirana i pla-zmidom pLS poznate koncentracije kako bi se odredila uspješnost transformacije. Iz nasumi-čno odabranih transformanata dobivenih transformacijom plazmidima pUC-GW-RadH i pUC-GW-NoxV izolirana je plazmidna DNA. Uspješnost izolacije ovih plazmida provjerena je a-garoznom gel elektroforezom, a izolirana DNA podvrgnuta je restrikcijskoj analizi kojom je potvrđeno da je bakterija E. coli uspješno transformirana plazmidima pUC-GW-RadH i pUC-GW-NoxV.

Abstract (english)

The aim of this study was to transform bacteria Escherichia coli with plasmids pUC-GW-RadH and pUC-GW-NoxV, and to confirm the transformation success by restriction analysis of plas-mid DNA isolated from selected transformants. The plasmids carry genes encoding for R-spe-cific alcohol-dehydrogenase and NADH-oxidase. Bacteria E. coli was additionally transformed with the plasmid pLS of known concentration to determine the transformation efficiency. Plas-mid DNA was isolated from randomly selected transformants obtained by transformation using plasmids pUC-GW-RadH and pUC-GW-NoxV. The success of the isolation was confirmed by agarose gel electrophoresis. The isolated plasmid DNA was subjected to restriction analysis, which confirmed that bacteria E. coli was successfully transformed with plasmids pUC-GW-RadH and pUC-GW-NoxV.
