Abstract

S obzirom na ubrzani porast industrijalizacije i potrebu za stvaranjem održivijih procesa, razvijen je koncept zelene kemije koji se oslanja na primjenu obnovljivih sirovina, alternativnih otapala i biokatalizatora za pretvorbu organskih spojeva u biotransformacijskim procesima. Stoga, u upotrebu sve više ulazi primjena eutektičkih otapala kao pogodnih reakcijskih medija za dobivanje enantiomerno čistih spojeva. Isto tako, provedena je enantioselektivna redukcija različito supstituiranih acetofenona u optički aktivne alkohole upotrebom bakterija mliječne kiseline Limosilactobacillus fermentum MC1 i Levilactobacillus brevis MB1, nakon čega je provedena optimizacija redukcije acetofenona kao modelnog supstrata i L. brevis MB1 kao pogodnog biokatalizatora u niskotemperaturnom eutektičkom otapalu. Biokatalitička redukcija acetofenona u otapalu betain : malična kiselina : glukoza s 30 %-tnim masenim udjelom vode rezultirala je većim enantiomernim viškom u odnosu na istu provedenu u hranjivoj podlozi. Abstract (english)

Considering the accelerated growth of industrialization and the need to create more sustainable processes, the concept of green chemistry was developed. It relies on the use of renewable raw materials, alternative solvents and biocatalysts for the conversion of organic compounds in biotransformation processes. More osmolyte-based deep eutectic solvents were prepared and their characteristic properties were determined by physico-chemical characterization. Likewise, the enantioselective reduction of differently substituted acetophenones into optically active alcohols was carried out using lactic acid bacteria Limosilactobacillus fermentum MC1 and Levilactobacillus brevis MB1, after which the optimization of the reduction of acetophenone as a model substrate and L. brevis MB1 as a suitable biocatalyst in a deep eutectic solvent was carried out. The biocatalytic reduction of acetophenone in the betaine : malic acid : glucose solvent with a 30% water content (w/w) resulted in a higher enantiomeric excess compared to the same one carried out in the nutrient medium.
