Abstract

Kadmij je teški metal koji iskazuje toksične učinke na ljudsko zdravlje kroz indukciju stanične proliferacije, apoptoze, oksidativnog stresa i oštećenja DNA. U ovom radu je istraţivan citotoksični i prooksidativni potencijal okolišno značajnih koncentracija kadmija te potencijalno mutageno djelovanje na humanim staničnim linijama. Iz dobivenih rezultata se moţe zaključiti da kadmij inducira staničnu proliferaciju i nastanak slobodnih radikala nakon 24 sata u rasponu koncentracija od 0,0017-0,44 nmol mL-1, koji odgovara koncentracijama kadmija izmjerenim u krvi djece i odraslih ljudi. Duţim izlaganjem, proliferativni i prooksidativni efekt se smanjuje, a povećava se citotoksični učinak niţih koncentracija kadmija. Pri tome su se stanice karcinoma debelog crijeva pokazale kao najosjetljivije. Ispitivanjem genotoksičnosti pokazalo se da kadmij uzrokuje pojavu točkastih mutacija, dok kromosomski lomovi nisu vidljivi.

Abstract (english)

Cadmium is a heavy metal that shows the toxic effects on human health through the induction of cell proliferation, apoptosis, oxidative stress and DNA damage. In this graduate work cytotoxic and prooxidative potential of environmentally relevant concentrations of cadmium and potentially mutagenic effects on the human cell lines was examined. From the results it can be concluded that cadmium induces cell proliferation and the formation of free radicals after 24 hours at the concentration range from 0,0017 to 0,44 nmol mL-1, which corresponds to the measured concentrations of cadmium in the blood of children and adults. With longer exposure on lower concentrations of cadmium, proliferative and prooxidative effect is reduced while the cytotoxic effect is increased. The colon cancer cells proved to be the most vulnerable on toxic effect of cadmium. Genotoxicity testing showed that cadmium caused point mutations, while the chromosomal breaks were not detected.
