Utjecaj teških metala na preživljavanje alge Euglena gracilis

Abstract

Industrijski i kućanski otpadi mogu izazvati zagađenje okoliša solima teških metala, a posljedično i promjene u biološkoj raznolikosti vodenih sustava. Alge su organizmi koji mogu u najvećoj mjeri apsorbirati i skladištiti teške metale. Kako su alge na donjoj razini vodenog hranidbenog lanca, najvažniji su čimbenik prijenosa zagađivača prema višim razinama. U ovom radu je proučavana toksičnost soli teških metala na algu E. gracilis, koja je poslužila kao biotestni mikroorganizam. Pokretljivost stanica, koncentracija pigmenata i sposobnost stvaranja biofilma E. gracilis proučavana je tijekom 12 dana uzgoja pri različitim koncentracijama soli teških metala (Cu2+, Pb2+, Co2+ and Hg2+ ;0.5 do 2.0 mM). Uočeno je da je fotosintetska učinkovitost i pokretljivost E. gracilis bila znatno smanjena nakon dodatka Co2+ iona, a potpuno nakon dodatka Hg2+ iona, dok Cu2+ i Pb2+ nisu izazvali inhibiciju pokretljivosti i fotosintetskih sposobnosti. Izloženost E. gracilis solima teških metala ukazala je na svojstvo ove alge da s vremenom kao odgovor na stres poboljša sposobnost stvaranja biofilma.

Abstract (english)

The pollution generated by heavy metals released by industrial and domestic sources cause serious changes in the aquatic ecosystem resulting in a loss of biological diversity. Algae are the main organisms that take up and store heavy metals. Since algae are at the bottom of the aquatic food chain, they are the most important vector for transfer of pollution to upper levels of the trophic chain in aquatic environments. The present study was aimed at the toxicity evaluation of heavy metals, using the flagellate Euglena gracilis as a biotest organism. Different parameters of the flagellate like motility, concentration of light harvesting pigments and biofilm formation were used as end points for the toxicity assessment, and observed results were calculated. E. gracilis was exposed for 12 days to Cu2+, Pb2+, Co2+ and Hg2+ from 0.5 to 2.0 mM. Hg2+ and Co2+ were found to be the most toxic of the investigated metals to photosynthetic efficiency and motility of E. gracilis , while Cu2+ and Pb2+ ions found to be the least toxic. Results showed that the exposure of E. gracilis to heavy metals stimulated the ability of this microorganism to increase the biofilm formation.
