Abstract

Celulaze (EC 3.2.1.4) kataliziraju hidrolizu β-1,4-D-glikozidnih veza u celulozi i imaju značajnu ulogu u prirodi reciklirajući ovaj polisaharid, koji je glavna komponenta stanične stijenke biljke. Ogroman potencijal koji celulaze imaju u biotehnologiji, pokretačka je snaga za stalna osnovna i primijenjena istraživanja ovih biokatalizatora iz gljiva i bakterija. Celulaze djeluju sinergistički s drugim hidrolitičkim enzimima kako bi se postigla potpuna razgradnja polisaharida do topljivih šećera, naime celobioze i glukoze, koje stanica zatim asimilira. Nekoliko vrsta plijesni proizvodi velike količine izvanstaničnih celulolitičkih enzima, dok bakterijski i nekoliko anaerobnih sojeva viših gljiva uglavnom proizvode celulolitičke enzime u kompleksu povezanom sa staničnom stijenkom koji se naziva celulosom. Celulaze i celulosomi imaju veliki potencijal za industrijsku primjenu i u središtu su napora da se stvori idealan mikroorganizam za pretvorbu celulozne biomase u vrijedne proizvode.

Abstract (english)

Cellulases (EC 3.2.1.4) catalyze the hydrolysis of β-1,4- D-glycosidic linkages in cellulose and play a significant role in nature by recycling this main polysaccharide component of the plant cell wall. The enormous potential cellulases have in biotechnology is the driving force for continuous basic and applied research on these biocatalysts from fungi and bacteria. Cellulases work in synergy with other hydrolytic enzymes to obtain the complete degradation of the polysaccharide to soluble sugars, namely cellobiose and glucose, which are then assimilated by the cell. Several fungi produce large amounts of extracellular cellulolytic enzymes, whereas bacterial and a few anaerobic fungal strains mostly produce cellulolytic enzymes in a complex associated with the cell wall called cellulosome. Cellulases and cellulosomes hold immerse potential for industrial application and are the focus
of much effort to engineer an ideal microorganism for converting cellulosic biomass to valuable products.
