Abstract

Biofarmaceutika je brzo rastuće područje unutar kojeg se provode istraživanja i koriste stanične linije, mikroorganizmi i njihovi proizvodi za terapijsku primjenu. Enzimi su izvanredni biokatalizatori koji ubrzavaju biokemijske reakcije. Posjeduju nekoliko specifičnih značajki kao što su velik katalitički potencijal, velika specifičnost za supstrat, kao i pH i temperaturni optimumi njihove katalitičke aktivnosti. Zbog ovih izvanrednih svojstava pronalaze različite kliničke primjene i sve se više koriste za liječenje širokog spektra bolesti, samostalno ili u kombinaciji s drugim terapijama, te čine važnu podskupinu modernih biofarmaceutika. Terapijska primjena enzima uključuje protuupalne agense, enzibiotike, fibrinolitičke agense, agense protiv tumora, probavne terapijske enzime, kao i enzime za rijetke bolesti. Razvoj novih terapijskih enzima kao i nadogradnja i poboljšanja postojećih primjenom proteinskog inženjerstva su prioritet. Izolaciju, regulaciju, proizvodnju i primjenu enzima kao biofarmaceutika treba unaprijediti do maksimalnog potencijala kako bi se iskoristile njihove prednosti u odnosu na kemijska terapijska sredstva u liječenju različitih bolesti.

Abstract (english)

Biopharmaceuticals is a fast emerging field that explores and employs biological organisms and their products for therapeutic applications. Enzymes are biocatalysts that accelerate the rate of biochemical reactions. They possess features like catalytic potential, substrate specificity as well as pH and temperature optima. Due to these attributes, they find diverse clinical applications, and are increasingly used to treat a wide range of diseases, alone or in combination with other therapies, so they are importan subclass of biopharmaceutics. Therapeutic applications of enzymes include anti‐ inflammatory agents, enzybiotics, fibrinolytic agents, anticancer agents, digestive therapeutic enzymes, as well as enzymes for rare diseases. Developement of new therapeutic enzymes as well as upgrading and improving existing ones using protein engineering are a priority. The isolation, regulation, production and application of microbial enzymes as biopharmaceuticals should be improved to the maximum potential in order to utilize their advantages over chemical therapeutic agents in the treatment of various diseases.
