Abstract

Genetička transformacija jest ulazak strane DNA u stanicu iz njene neposredne okoline. Razlikujemo više metoda transformacije poput kemijske metode, biolističke, transformacije pomoću ultrazvuka (sonoporacije), „tribos“ transformacije i elektrotransformacije (elektroporacije). U ovom radu detaljnije je opisana metoda elektroporacije, transformacija koja se temelji na primjeni kratkog pulsa visokog napona. Elektroporacija je jedna od najučinkovitijih metoda transformacije kod bakterija Escherichia coli. Cilj ovog rada bio je pripremiti elektrokompetentne stanice bakterije Escherichia coli soja MC1061 te provjeriti kompetentnost pripremljenih stanica transformacijom s plazmidom pLS42.

Abstract (english)

Genetic transformation involves the entry of foreign DNA into the cell. We distinguish several transformation methods such as chemical method, biolistic method, sonic transformation (sonoporation), “tribos” transformation and electrotransformation (electroporation). Electrotransformation, transformation based on the application of a short puls of high voltage is described in more detail in this work. Electroporation is one of the most effective transformation methods when it comes to Escherichia coli. The aim of this study was to prepare electrocompetent cells of Escherichia coli strain MC1061 and to check the competence of the prepared cells by transformation with plasmid pLS42.
