Abstract

Tehnologija rekombinantne DNA je skup molekularno - genetičkih metoda koje omogućuju uvođenje preciznih genetičkih promjena u živu stanicu ili organizam u svrhu konstrukcije rekombinantnih molekula DNA. Primjena ove tehnologije u biologiji i srodnim znanostima rezultirala je mnogim revolucionarnim otkrićima. U ovom radu spomenutim metodama konstruiran je plazmid pBAD – NoxV. Plazmid je konstruiran ugradnjom fragmenta (inserta) DNA izdvojenog iz plazmida pUC – GW – NoxV, koji sadrži gen NoxV Ecoli Opti, u okosnicu plazmida pBADHHEC koji je korišten kao vektor te sadrži gene za rezistenciju na antibiotik ampicilin, ishodište replikacije u bakteriji Escherichii coli, gen araC i promotor araBAD. Iz odabranih transformanata bakterije E. coli izolirana je plazmidna DNA čija je struktura provjerena restrikcijskom analizom. Rezultati analize su potvrdili uspješnu konstrukciju
plazmida pBAD - NoxV.

Abstract (english)

Recombinant DNA technology is a set of molecular-genetic methods that enable precise genetic changes to be introduced into a cell or organism for the purpose of constructing recombinant DNA molecules. The application of this technology in biology and related sciences has resulted in many revolutionary discoveries. In this work, the plasmid pBAD – NoxV was constructed using by mentioned methods. The plasmid was constructed by incorporating a fragment (insert) of DNA isolated from the plasmid pUC - GW - NoxV containing the NoxV Ecoli opti gene into the backbone of the plasmid pBADHHEC, which was used as a vector and contains genes for resistance to the antibiotic ampicillin,
the origin of replication in the Escherichia coli bacterium, the araC gene and the araBAD promoter. From the selected transformants, plasmid DNA was isolated and subjected to restriction analysis. The result of the analysis confirmed the successful construction of the plasmid pBAD - NoxV.
