Djelovanje hidrolizata proteina lana tijekom induciranog oksidacijskog stresa u CHO DP-12 stanicama

Abstract

Pogača lana bogata je proteinima, a industrijski je slabo iskorištena. U posljednje vrijeme postaje sve zanimljivija za istraživanja, što zbog bogatstva bioaktivnim peptidima s potencijalnim antioksidativnim djelovanjem, tako i zbog prisustva ostalih bioaktivnih komponenata. S obzirom na navedeno, hidrolizati proteina pogače lana imaju potencijal primjene u kulturama životinjskih stanica kao dodatak medijima za uzgoj. Pri tome se koriste kao zamjena za dio seruma, ili pak samo kao dodatan sastojak koji zbog bioaktivnih svojstava može blagotvorno djelovati na stanice u kulturi. U ovom radu cilj je bio ispitati mogu li pripremljeni hidrolizati proteina iz pogače lana imati protektivno djelovanje na CHO DP-12 stanice tijekom induciranog oksidativnog stresa. Tijekom eksperimenata, stanice su nacijepljene u mediju za uzgoj s dodatkom različitih koncentracija hidrolizata proteina lana u rasponu 0,1-5 g/L. Stanice su brojane metodom Trypan Blue, vijabilnost im je određivana MTS metodom, a dobiveni podaci statistički su obrađeni. Provedbom eksperimenta, došlo se je do zaključka da hidrolizat lana dobiven enzimom neutraza i dodan u medij u koncentraciji od 0,5 g/L pokazuje najjače protektivno djelovanje na stanice CHO DP-12 izložene oksidacijskom stresu, dok hidrolizat lana dobiven enzimom alkalaza i u medij dodan u koncentraciji od 0,1 g/L pokazuje blago inhibirajući učinak na stanice izložene oksidacijskom stresu. Međutim, da bi se ovi zaključci potvrdili, potrebno je napraviti još istraživanja na istu temu jer je radova za usporedbu u literaturi vrlo malo.

Abstract (english)

Flax cake is rich in proteins, but industrially it has been little used. Recently, it has become more and more interesting for research, due to the wealth of bioactive peptides with potential anti-oxidative effect, as well as the presence of other bioactive components. Considering the above, flax cake protein hydrolysates have the potential to be used in animal cell cultures as an addition to culture media. In doing so, they are used as a substitute for part of the serum, or simply as an additional ingredient that, due to its bioactive properties, can have a beneficial effect on cells in culture. In this work, the aim was to examine whether prepared protein hydrolysates from flax cake can have a protective effect on CHO DP-12 cells during induced oxidative stress. During the experiments, cells were inoculated in culture medium supplemented with different concentrations of flax protein hydrolysates in the range 0.1-5 g/L. Cells were counted using the Trypan Blue method, their viability was determined using the MTS method, and the obtained data were statistically processed. Through the experiment, it was concluded that flax hydrolysate obtained with the enzyme neutrase and added to the
medium at a concentration of 0.5 g/L shows the greatest protective effect on CHO DP-12 cells exposed to oxidative stress, while flax hydrolysate obtained with the enzyme alcalase and added to the medium at a concentration of 0.1 g/L, shows a slightly inhibiting effect on cells exposed to oxidative stress. However, in order to confirm these conclusions, it is necessary to do more research on the same topic because there are very few works available for comparison in the literature.
