Abstract

Ljubičaste nesumporne bakterije su fotosintetski prokarioti koji svjetlosnu energiju pretvaraju u kemijsku procesom anoksigene fotosinteze. Najčešće se izoliraju iz otpadnih voda i ekstremnih staništa. Njihova prisutnost dovodi do biorazgradnje organskih i anorganskih spojeva, kao i do biosinteze visokovrijednih nusprodukata. Biomasa ljubičastih nesumpornih bakterija sadrži visoku koncentraciju proteina, esencijalnih aminokiselina, pigmenata i vitamina. Najvažnija primjena ove skupine bakterija je u obradi otpadnih voda prehrambene industrije te u biotehnološkoj proizvodnji biopolimera. U ovom radu su određene morfološke i fenotipske karakteristike dviju vrsta ljubičastih nesumpornih bakterija-Rhodobacter capsulatus JCM 21090 i Rhodobacter azotoformans JCM 9340. Rezultati istraživanja pokazali su da se radi o gram-negativnim, pokretnim bakterijama, koje prilikom rasta na krutoj hranjivoj podlozi stvaraju glatke okrugle kolonije. Tijekom duljeg uzgoja u tekućoj hranjivoj podlozi došlo je do sinteze pigmenta i obojenja podloge u crvenu boju. Fenotipskom identifikacijom koristeći SDS elektroforezu, dobiven je prikaz ukupnih i površinskih proteina, koji se međusobno razlikuju ovisno o periodu inkubacije bakterija. Dobiveni rezultati daju važan doprinos za buduće karakterizacije i primjenu ljubičastih nesumpornih bakterija.

Abstract (english)

Purple non-sulfur bacteria are photosynthetic prokaryotes that convert light energy into the chemical energy through the process of anoxygenic photosynthesis. They are most often isolated from wastewater and extreme habitats. Their presence leads to the biodegradation of organic and inorganic compounds, as well as to the biosynthesis of high-value byproducts. The biomass of purple non-sulfur bacteria contains a high concentration of proteins, essential amino acids, pigments and vitamins. The most important use of this type of bacteria is in the treatment of wastewater from the food industry and in the biotechnological production of biopolymers. In this research, the morphological and phenotypic characteristics were determined of two strains of purple non-sulfur bacteria-Rhodobacter capsulatus JCM 21090 and Rhodobacter azotoformans JCM 9340. The results of the research showed that they are gram-negative, motile bacteria which create smooth round colonies when growing on a solid nutrient substrate. During the long cultivation in the liquid nutrient medium, pigment synthesis occurred and the medium was colored red. By phenotypic identification using SDS electrophoresis, a display of total and surface proteins was obtained, which differ from each other depending on the period of bacterial incubation. The obtained results provide an important contribution to the future characterization and application of purple non-sulfur bacteria.
