Abstract

Enzimski potpomognute ekstrakcije su metode koje se temelje na korištenju enzima koji hidroliziraju staničnu stijenku biljaka, kako bi se povećala efikasnost ekstrakcijskog predtretmana te se smanjila količina organskog otapala koje se koristi tijekom ekstrakcije. U ovom radu određivana je aktivnosti enzima celulaze, ksilanaze i pektinaze. Određivanje je provedeno prema Kracherovoj metodi. To je kolorimetrijska metoda u kojoj DNSA uz prisutnost reducirajućih šećera prelazi u 3-amino-5-nitrosalicilnu kiselinu, te se karbonilna skupina šećera reducira u karboksilnu skupinu. U usporedbi s rezultatima određivanja aktivnosti celulaze u otopinama supstrata celuloze i lista masline, možemo zaključiti da je aktivnost veća ako je supstrat čist. Također na prinos ekstrakcije utječu i temperatura, pH, vrijeme ekstrakcije, količina enzima i vrsta enzima. Korištenjem enzima celulaze značajno se povećao prinos ekstrakcije fenola iz lista masline.

Abstract (english)

Enzyme-assisted extractions are methods based on the use of enzymes that hydrolyze plant cell walls, in order to increase the efficiency of the extraction pretreatment and reduce the amount of organic solvent used during extraction. In this thesis, cellulase, xylanase and pectinase enzyme activity was determined. The determination was carried out according to Kracher's method. It is a colorimetric method in which, in the presence of reducing sugars, DNSA turns into 3-amino-5-nitrosalicylic acid, and the carbonyl group of the sugar is reduced to a carboxyl group. In comparison with the results of the determination of cellulase activity in cellulose and olive leaf substrate solutions, we can conclude that the activity is higher if the substrate is clean. Although, extraction yield is also affected by temperature, pH, extraction time, amount of enzyme and type of enzyme. Utilization of cellulase enzyme significantly increased the yield of phenols extraction from olive leaves.
