Abstract

Adjuvanti su komponente koje se dodaju u cjepiva s ciljem razvoja snažnog i dugotrajnog imunosnog odgovora na cjepni antigen. Nastojanja za pronalaženjem učinkovitog i sigurnog adjuvanta važna su i zbog podjediničnih cjepiva slabije imunogeničnosti. Razvoj nove generacije adjuvanata često uključuje različite nanomaterijale za produženo otpuštanje antigena, a koji istovremeno mogu i sami posjedovati imunostimulirajuća svojstva. Supramolekulski samoorganizirani peptidni hidrogelovi imaju veliki potencijal za primjenu u isporuci lijekova i cjepiva zbog svoje biokompatibilnosti. U okviru ovoga diplomskog rada istražena su adjuvantna svojstva supramolekulskih sustava temeljenih na samoorganiziranim hidrogelovima tripeptida Ac-L-Phe-L-Phe-L-Ala-NH₂; Ac-L-Phe-L-Phe-β-AlaOMe i Ac-L-Phe-L-Phe-L-Ala-COOH. Također istraženi su i nanokompoziti hidrogelatora Ac-L-Phe-LPhe-L-Ala-NH₂ i polimera, polietilen glikola (PEG) i polivinil alkohola (PVA). U hidrogelove je ugrađen modelni antigen, ovalbumin (OVA) i ELISA testom je određena količina nastalih specifičnih imunoglobulina IgG. Izmjerene su količine nastalih imunoglobulina anti-ovalbumin IgG/IgG1/IgG2a i pokazano je da Ac-L-Phe-L-Phe-L-Ala-NH₂ značajnopojačava imunoreakciju, dok Ac-L-Phe-L-Phe-βAla-OMe i Ac-L-Phe-L-Phe-L-Ala-COOH utječu na smjer imunoreakcije.

Abstract (english)

Adjuvants are components added to vaccines in order to develop a strong and long-lasting immune response to the vaccine antigen. Efforts to find an effective and safe adjuvant are important because of the lower immunogenicity of subunit vaccines. Development of new generation of adjuvants often includes different nanomaterials for sustained release of antigen and at the same time they may possess immunostimulating properties on their own. Supramolecular self-assembled peptide hydrogels have a great potential for applications in drug and vaccine delivery, and it has been shown that certain peptide hydrogelators can enhance and modulate the immune response. In the frame of this graduate thesis the adjuvant properties of supramolecular hydrogel based on tripeptide Ac-L-Phe-L-Phe-L-Ala-NH₂, Ac-LPhe-L-Phe-β-Ala-OMe and Ac-L-Phe-L-Phe-L-Ala-COOH were tested. Nanocomposite hydrogels of Ac-L-Phe-L-Phe-L-Ala-NH₂ with poly(ethylene glycol) and poly(vinyl alcohol) were also tested. A model antigen, ovalbumin, was incorporated into the hydrogels, and the amount of specific antiovalbumin IgG/IgG1/IgG2a immunoglobulins was determined. It was shown that Ac-L-Phe-L-Phe-LAla-NH₂ significantly enhances the immunoreaction, while Ac-L-Phe-L-Phe-β-Ala-OMeand Ac-L-PheL-Phe-L-Ala-COOH shift the immune response towards the Th-1 type of immune reaction.
