Abstract

U ovom radu provedena je analiza modela metabolizma kolesterola u računalnom programu CellDesigner. Simulacije modela provedene su koristeći različite početne vrijednosti kolesterola iz hrane koje su dobivene iz jelovnika izrađenih u okviru ovog rada – jelovnik za vegetarijansku prehranu, uravnoteženu prehranu i ketogenu prehranu. Model uspješno simulira promjenu koncentracije kolesterola i njegovih metabolita. Rezultati ukazuju na utjecaj mase kolesterola iz hrane na masu crijevnog i jetrenog kolesterola te na manjak utjecaja na koncentracije metabolita kolesterola u krvnoj plazmi. Provedena je analiza lokalne parametarske osjetljivosti u kojoj se crijevni kolesterol pokazao kao najosjetljivija varijabla, a parametri jetrene sinteze nascentnog HDL-a i nastanka VLDL kolesterola iz jetrenog kolesterola pokazali su najveći utjecaj na izlazne rezultate nakon simulacije modela.

Abstract (english)

Analysis of cholesterol metabolism model was performed using the CellDesigner software. Simulations of the constructed model were conducted using different initial values of dietary cholesterol, obtained from three different menus created for the purposes of thesis – vegetarian diet menu, balanced diet menu and keto diet menu. Model successfully simulates metabolism of cholesterol and its metabolites. Results show that mass of dietary cholesterol has an impact on masses of intestinal and hepatic cholesterol, but not on the concentrations of cholesterol metabolites in the blood plasma. Based on the analysis of local parameter sensitivity it was concluded that intestinal cholesterol is the most sensitive variable of the model. Parameters defining hepatic nascent HDL synthesis and VLDL cholesterol formation were shown to have the greatest impact on the results of the simulation.
