Abstract

Mnogi mikroorganizmi mogu koristiti ugljikovodike kao izvor ugljika i energije za metaboličke aktivnosti te su takvi mikroorganizmi široko rasprostranjeni u prirodi. Glavni mikrobiološki problem u naftnoj industriji su kontaminacije uskladištenih produkata odnosno goriva, koje uzrokuje gubljenje kvalitete proizvoda te stvaranja mulja što dovodi do kvarenja proizvoda, začepljena cjevovoda i tankova. Ugljikovodici iz goriva su čest uzrok onečišćenja okoliša pa na naftnoj i industriji leži velika odgovornost za rješavanje problema onečišćenja okoliša. S ciljem identifikacije mikroorganizama, u ovom radu izolirani su mikroorganizmi iz spremnika s dizelskim gorivom. Korištena je metoda identifikacije bazirana na proteomskom pristupu koja uključuje primjenu spektrometra masa MALDI/TOF-TOF. Od 9 izoliranih vrsta, biotipizacijom pomoću programskog alata Protein Reader identificirano je 5 različitih vrsta. Najzastupljenija vrsta je Nectria haematococca.

Abstract (english)

Many microorganisms can utilize hydrocarbons as sole sources of carbon as energy for metabolic activities and these microorganisms are present and widely distributed in the nature. The major microbial problem in the petroleum refining industry is contamination of stored products, which can lead to loss of product quality, formation of sludge and deterioration of pipework and storage tanks. Hydrocarbons from fuel are common cause of environmental pollution so there is a great responsibility on petroleum industry for solving the problem of hydrocarbons pollution. In order to identify microorganisms, in this work we isolated microorganisms from diesel fuel storage tank. Method used for identification is based on proteomic approach and includes application of mass spectrometry. From 9 isolated species we identified 5 different species by biotypization protocol using software Protein Reader. Most common species is Nectria haematococca.
