Abstract

Mnoge supstance, uključujući teške metale, koje se koriste u različitim područjima u industriji i/ili poljoprivredi, djeluju kao inhibitori enzima, koji posljedično tome ne mogu vezati supstrat. Iako nije jako osjetljiva, metoda korištenja biosenzora za detekciju tragova teških metala ima dinamičan trend rasta i široko se primjenjuje za poboljšanje „kvalitete života“, zbog osjetljivosti, selektivnosti i jednostavnosti samog biosenzora. U ovom istraživanju, korišteni su enzimski biosenzori, među kojima se biosensor temeljen na ureazi pokazao kao vrlo jednostavan, osjetljiv na tragove teških metala, stabilan i brz. Određen je i tip inhibicije te utvrđeno da teški metali inhibiraju ureazu na nekompetitivan način. Također, određena je i učinkovitost teških metala kao inhibitora ureze. Cu2+, Hg2+ and Ag+ su pokazali najjači inhibitorni učinak. Naposljetku, određena je i granica detekcije uređaja.

Abstract (english)

Many compounds, including heavy metals, used in different fields of industry and/or agriculture act as inhibitors of enzymes, which, as a consequence, are unable to bind the substrate. Even if it is not so sensitive, the method for detecting heavy metal traces using biosensors has a dynamic trend and is largely applied for improving the “life quality”, because of the biosensor’s sensitivity, selectivity, and simplicity. In this research, enzyme-based biosensors were used, among which urease-based biosensor proved itself as very simple, sensitive on heavy metal traces, stable and fast. The inhibition type was examined and established that urease is inhibited by heavy metals in non-competitive manner. Also, the effectiveness of the heavy metal ions as inhibitors of urease was determined. Cu2+, Hg2+ and Ag+ were the ones with the highest inhibitory effect. Finally, the device’s limit of detection was also determined.
