Abstract

Peptidomimetici su spojevi koji svojom trodimenzijskom strukturom oponašaju sekundarne strukturne elemente prirodnih peptida i proteina, a odlikuju se boljom biodostupnošću i proteolitičkom stabilnošću. Umetanjem 1,1'-disupstituiranih ferocenskih kalupa u peptidni lanac inducira se tvorba strukture okreta kao i struktura nalik β-nabranim pločama. Ferocen1,1'-diamin kao rigidni kalup omogućava tvorbu intralančanih vodikovih veza kojima se zatvara 14-člani prsten karakterističan za prirodne β-nabrane ploče. Konjugati ferocen-1,1'-diamina s hidrofobnim aminokiselinama valinom (1,4), leucinom(2,5) te fenilalaninom (3, 6) podvrgnuti su testiranju biološke aktivnosti na zdravoj HFF-1 i tumorskoj MCF-7 staničnoj liniji. Testovi su provedeni u in vitro uvjetima pomoću standardizirane MTT metode. Najviši stupanj toksičnosti i sposobnost raspoznavanja tumorskih od zdravih stanica pokazali su L-konjugati ferocen-1,1'-
diamina s leucinom (2) i fenilalaninom (3).

Abstract (english)

Peptidomimetics are compounds whose three-dimensional structure mimics the secondary structural elements of natural peptides and proteins. They are characterized by better bioavailability and proteolytic stability. The insertion of a 1,1-disubstituted ferrocene scaffold into a peptide chain induces the formation of a turn and β-sheet-like structure. Ferrocene-1,1'-diamine as a rigid scaffold enables the formation of intrachain hydrogen bonds that allow the formation of 14-membered ring common to native β-sheets. The conjugates of ferrocene-1,1'-diamine with the hydrophobic amino acids valine (1,4), leucine (2,5), and phenylalanine (3,6) were tested for their biological activity on healthy HFF-1 and tumor MCF-7 cell lines. The tests were performed under in vitro conditions using the standardized MTT method. The L-conjugates of ferrocene-1,1'-diamine with the leucine (2) and phenylalanine (3) showed the highest degree of toxicity and the ability to differentiate between tumor and healthy cells.
