Abstract

Zbog važnosti staničnih nosača u različitim granama znanosti, medicine i industrije došlo je do porasta interesa za pronalaskom nosača koji bi se mogli izraditi jednostavnim metodama korištenjem jeftinih sirovina. Pritom je velika pozornost posvećena hidrogelovima budući da pokazuju iznimno dobra fizikalno–kemijska i biološka svojstva koja ih čine potencijalno superiornima u odnosu na druge oblike nosača. Cilj ovog rada bila je izrada hidrogelova od pektina i proteina lana, uz dodatak hidrolizata kolagena, koji bi se potencijalno mogli koristiti kao stanični nosači. Hidrogelovi su dobiveni termičkom obradom otopine pektina (5 %, w/v) s dodatkom proteina lana i/ili hidrolizata kolagena (5,3 %, w/v), uz naknadno geliranje pomoću kalcijevih iona. Rezultati analize izrađenih gelova pokazali su da dodatak proteina lana i hidrolizata kolagena ima negativan utjecaj na mehanička svojstva hidrogelova, ubrzava njihovo isušivanje i usporava bubrenje, a nema utjecaja na citoadhezivna svojstva. Izrađeni hidrogelovi nisu citotoksični, ali se zbog loših citoadhezivnih svojstava ne mogu koristiti kao stanični nosači.

Abstract (english)

Due to the importance of cell scaffolds in various branches of science, medicine and industry, there has been a growing interest in finding scaffolds that could be made using simple methods and cheap raw materials. Great attention is paid to hydrogels since they exhibit exceptionally good physico-chemical and biological properties that make them potentially superior to other forms of scaffolds. The aim of this study was to produce pectin and flaxseed protein hydrogels, with the addition of collagen hydrolysate, which could potentially be used as cell scaffolds. The hydrogels were made by thermal treatment of pectin solutions (5 %, w/v), with addition of flaxseed proteins and/or collagen hydrolysate (5.3 %, w/v), and by subsequent gelling using calcium ions. The results of the analysis of the made gels showed that the addition of flaxseed protein and collagen hydrolysate has a negative effect on the mechanical properties of hydrogels, accelerates their drying, slows swelling, and has no effect on cell-adhesive properties. The produced hydrogels are not cytotoxic, but due to poor celladhesive properties, they cannot be used as cell scaffolds.
