Abstract

Konjugacijom organometalnog spoja ferocena s biomolekulama nastaju kemijski raznoliki spojevi koje odlikuje iznimna stabilnost, netoksičnost i povećana bioaktivnost. U ovom je radu ispitana in vitro biološka aktivnost osam ferocenskih konjugata s homo- i heterokiralnim dipeptidom pripravljenim iz alanina (Ala) i prolina (Pro) na humanoj tumorskoj staničnoj liniji HeLa. Antitumorski učinak je ispitan određivanjem stanične vijabilnosti te klonogenom analizom. Protočnom citometrijom određeno je dolazi li do aktivacije procesa stanične smrti. Ispitivani ferocenski peptidomimetici imaju inhibitorni učinak na rast HeLa stanične linije. Najjače citotoksično djelovanje pokazuju spojevi 1, 2 i 5. Inhibitorno djelovanje spojeva 1 i 2 može se povezati s poticanjem stanične smrti apoptozom. Najizraženiji antitumorski potencijal pokazuju spojevi koji sadrže tert-butoksikarbonilnu (Boc)-zaštitnu skupinu te L-Pro, odnosno spojevi 1 i 2.

Abstract (english)

By conjugating the organometallic compound ferrocene with biomolecules, chemically diverse compounds are formed, characterized by exceptional stability, non-toxicity, and increased bioactivity. This study examined the in vitro biological activity of eight ferrocene conjugates with homo- and heterochiral dipeptide formed of alanine (Ala) and proline (Pro) on the human tumor cell line HeLa. The antitumor effect was examined by determining cell viability and performing a clonogenic assay. Flow cytometry was used to determine whether the process of cell death is activated. The investigated ferrocene peptidomimetics have an inhibitory effect on the growth of the HeLa cell line. Compounds 1, 2, and 5 show the strongest cytotoxic activity. The inhibitory effect of compounds 1 and 2 can be linked to the induction of cell death by apoptosis. The compounds containing the tert-butoxycarbonyl (Boc)-protective group and L-Pro, specifically compounds 1 and 2, show the most pronounced antitumor potential.
