Abstract

Fluorescentna in situ hibridizacija (eng. fluorescent in situ hybridization, FISH) je učinkovita vizualna tehnika koja omogućuje identifikaciju i lokalizaciju određenih sekvenci DNA na kromosomima. Ovaj teorijski završni rad bavi se tehnikom FISH, njezinim povijesnim razvojem, primjenom u molekularnoj genetici i genetičkom inženjerstvu, te mogućnostima i ograničenjima pojedinih varijanti ove metode. Rad započinje kratkim povijesnim pregledom razvoja metoda za vizualizacije kromosoma i citogenetičkih postupaka, te razvojem samog FISH-a. Nadalje, u ovom radu opisane su različite varijacije ove metode, uključujući tzv. Multipleks FISH (eng. Multiplex Fluorescent in situ Hybridization, M-FISH) Fibre-FISH i interfazni FISH, te i njihove prednosti i ograničenja. Opisane su i kliničke primjene FISH-a, prvenstveno u dijagnostici nasljednih bolesti poput kronične mijeloične leukemije (eng. Chronic Myeloid Leukemia, CML) te razmatrane mogućnosti daljnjeg napretka u FISH tehnologiji.

Abstract (english)

Fluorescent in situ hybridization (FISH) is an effective visual technique that enables the identification and localization of specific DNA sequences on chromosomes. This theoretical thesis focuses on the FISH technique, its historical development, application in molecular genetics and genetic engineering, and the possibilities and limitations of certain variants of this method. The thesis first brings a brief historical overview of the development of methods for chromosome visualization and cytogenetic procedures, as well as the development of FISH itself. Furthermore, this paper describes various variations of this method, including the so-called Multiplex FISH (eng. Multiplex Fluorescent in situ Hybridization, M-FISH) Fibre-FISH and interphase FISH, and their advantages and limitations. Clinical applications of FISH are also described, primarily in the diagnosis of hereditary diseases such as Chronic Myeloid Leukemia (CML), and possibilities for further progress in FISH technology are discussed.
