Abstract

Ferocenski konjugati su organometalni spojevi sa specifičnim elektrokemijskim i biološkim svojstvima, što ih čini zanimljivima za istraživanje u razvoju novih lijekova. Budući da mnogi od njih pokazuju antitumorski učinak, u ovom radu je ispitana in vitro aktivnost tri biokonjugata ferocena i 2 tiouracila na HeLa stanicama praćenjem stanične vijabilnosti te klonogenom analizom. Nadalje, protočnom citometrijom određeno je s kojim se staničnim procesom može povezati zapažena citotoksičnost. Ispitani biokonjugati ferocena i 2-tiouracila djeluju inhibitorno na rast HeLa stanica pri čemu je inhibitorni učinak ovisan o dozi. Najsnažniji citotoksični učinak pokazuje spoj 1 sa IC₅₀ vrijednošću 23,39 μM. Inhibitorni učinak može se povezati s poticanjem stanične smrti mehanizmom apoptoze. Slijedom navedenog može se zaključiti da biokonjugati ferocena i 2-tiouracila imaju potencijal kao antitumorski agensi.

Abstract (english)

Ferrocene conjugates are organometallic compounds with specific electrochemical and biological properties, what makes them interesting in drug development research. Since many of them exhibit antitumor effects, in this study the in vitro activity of three ferrocene and 2-thiouracil bioconjugates on HeLa cells was examined by cell viability and clonogenic assays. Furthermore, flow cytometry was used to determine which cellular processes are associated with the observed cytotoxicity. The tested bioconjugates of ferrocene and 2-thiouracil have an inhibitory effect on the growth of HeLa cells, which is dose-dependent. The strongest cytotoxic effect is shown by compound 1 with an IC₅₀ value of 23.39 μM. The inhibitory effect can be related to the induction of cell death by the apoptosis. Therefore, it can be concluded that bioconjugates of ferrocene and 2-thiouracil have potential as antitumor agents.
