Abstract

Svrha ovog rada bila je identificirati i kvantificirati prisutnost bioaktivnih peptida u nusproizvodima mliječne industrije koji obuhvaćaju slatku i kiselu sirutku te permeat. Za provođenje identifikacije bioaktivnih peptida korištena je spregnuta tehnika tekućinske kromatografije obrnutih faza (nano LC) sa spektrometrijom masa (MS). Dobivene proteinske sekvence peptida pretražene su pomoću baze podataka za bioaktivne peptide izolirane iz mlijeka (engl. Milk Bioactive Peptide Database). Neciljanom i podatkovno ovisnom analizom utvrđena je prisutnost čak 10 bioaktivnih peptida u analiziranim uzorcima, a to su SQSKVLPVPQ, TQTPVVVPPFLQPE, QEPVLGPVRGPFPIIV, VLGPVRGPFP, RDMPIQAF, EPVLGPVRGPFP, VYPFPGPIPN, SQSKVLPVPQKAVPYPQ, MPFPKYPVEP i YQEPVLGPVR. Za ove peptide su u znanstvenoj literaturi potvrđena i opisana bioaktivna svojstva poput antioksidativnog, antitrombotičkog, imunomodulatornog i ACE-inhibitornog (antihipertenzivno). Kvantifikacija identificiranih peptida je provedena pomoću neciljane SWATH-DIA (engl. Sequential Window Acquisition of all Theoretical Mass Spectra-Dana Independent Acquisition) analize. Rezultati su statistički obrađeni pomoću web platforme MetaboAnalyst verzija 6.0. Utvrđeno je kako je većina identificiranih bioaktivnih peptida statistički značajnije zastupljena u uzorcima kisele sirutke.

Abstract (english)

The purpose of this study was to identify and quantify the presence of bioactive peptides in by-products of the dairy industry, specifically sweet whey, acid whey, and permeate. Identification of bioactive peptides was carried out using a coupled technique of reverse-phase liquid chromatography (nano LC) with mass spectrometry (MS). The peptide sequences obtained were searched against the Milk Bioactive Peptide Database. Through untargeted and data-dependent analysis, the presence of ten bioactive peptides was confirmed in the analyzed samples, including SQSKVLPVPQ, TQTPVVVPPFLQPE, QEPVLGPVRGPFPIIV, VLGPVRGPFP, RDMPIQAF, EPVLGPVRGPFP, VYPFPGPIPN, SQSKVLPVPQKAVPYPQ, MPFPKYPVEP, and YQEPVLGPVR. These peptides have been confirmed and described in scientific literature for bioactivities such as antioxidant, antithrombotic, immunomodulatory, and ACE-inhibitory (antihypertensive) properties. Quantification of the identified peptides was performed using an untargeted SWATH-DIA (Sequential Window Acquisition of all Theoretical Mass Spectra-Data Independent Acquisition) analysis. Statistical processing of the results was conducted using the MetaboAnalyst web platform, version 6.0. It was found that most of the identified bioactive peptides were significantly more abundant in acid whey samples.
