Abstract

U ovom je radu proučavana učestalost pojave gubitka heterozigotnosti gena TP53 kod osoba koje boluju od meningeoma. U tu svrhu, korištena metoda bila je analiza polimorfizma u duljini restrikcijskih fragmenata. Kao uzorak korištena je genomska DNA iz meningeoma i iz leukocita. Najprije je umnožen dio introna 6 gena TP53 PCR-om iz uzoraka, a zatim je vršena restrikcija PCR produkta endonukleazom MspI. Rezultati su bili prikazani elektroforezom na agaroznom gelu. Samo je 21,05% pacijanata u ovom uzorku bilo informativno za ovaj genski marker. U niti jednog pacijenta nije pronađen gubitak heterozigotnosti. Međutim, pošto je uzorak heterozigotnih pacijenata malen, bilo bi dobro istražiti ovu pojavu na većem broju heterozigotnih pacijanata.

Abstract (english)

This paper studied the incidence of loss of heterozygosity of TP53 gene in patients suffering from meningioma. For this purpose, the method used was the Restriction Fragment Length Polymorphism analysis on samples of genomic DNA from meningioma and leukocytes. First, part of intron 6 of gene TP53 was amplified by PCR, and than the restriction of PCR products was performed with MspI endonuclease. Results were obtained by electrophoresis on agarose gel. Only 21,05% of patients were informative for this genetic marker. No patient showed loss of heterozygosity. However, since the sample of heterozygous patients is small, it is necessary to investigate this occurrence on a larger number of heterozygous patients.
