Abstract

Activities related to petroleum industry often lead to hydrocarbon contamination of soil and water. For many years, mechanical and chemical methods have been the only way for removing hydrocarbons from contaminated sites. Since these methods often have limited effectiveness and can be very expensive, the aim is to replace them with more efficient and cost-effective methods, such as microbial degradation by indigenous microorganisms in water and soil capable of degrading hydrocarbon contaminants.
In this work, metagenome from oil-polluted sea sediments was analyzed using bioinformatic methods with the aim of assessing the biopotential of microbial communities for remediation of hydrocarbon contamination. Targets were enzymes involved in aerobic degradation of short- and long-chain alkanes as well as enzymes for anaerobic alkane degradation.
Analyzed metagenome contained enzymes both for aerobic and anaerobic alkane degradation, with majority of genes of interest belonging to alkB alkane hydroxylase and cytochrome P450 alkane hydroxylase families.
Selected complete genes from every family of interest will be sent for synthesis, expressed and characterized with the final goal of assessing their potential for bioremediation of oil-polluted sea and soil.

Abstract (french)

Les activités liées à l'industrie du pétrole conduisent souvent à la contamination aux hydrocarbures des sols et de l'eau. Pendant de nombreuses années, des méthodes mécaniques et chimiques ont été le seul moyen pour éliminer les hydrocarbures provenant de sites contaminés. Puisque ces méthodes ont souvent une efficacité limitée et peuvent être très coûteux, l'objectif est de les remplacer par des méthodes plus efficaces et plus rentables, tels que la dégradation microbienne de micro-organismes indigènes dans l'eau et le sol capables de dégrader les contaminants d'hydrocarbures. Dans ce travail, métagénome de sédiments marins huile polluée a été analysée en utilisant des méthodes bioinformatiques dans le but d'évaluer la biopotentiel des communautés microbiennes pour l'assainissement de la contamination aux hydrocarbures. Les cibles étaient des enzymes impliquées dans la dégradation aérobie des alcanes à court et à longue chaîne ainsi que des enzymes pour la dégradation anaérobie de l'alcane. Métagénome analysé contenait deux enzymes de la dégradation des alcanes aérobie et anaérobie, à la majorité des gènes d'intérêt appartenant à alkB alcane hydroxylase et le cytochrome P450 familles alcane hydroxylase. Gènes complets sélectionnés de chaque famille d'intérêt seront envoyés pour la synthèse, exprimés et caractérisés par le but final de l'évaluation de leur potentiel de biodégradation de la mer et de la terre huile polluée.
