Utjecaj S-nitrozilacije transkripcijskog faktora LuMYB61 na biosintezu lignana u lanu

Abstract

In this study we investigate the role of a MYB family transcription factor LuMYB61. LuMYB61 contains highly conserved R2R3 domain and has a supposed role in lignan biosynthesis. Pinoresinol lariciresinol reductase 1, encoded by the LuPLR1 gene in Linum usitatissimum L. is responsible for the biosynthesis of flax’s most abundant lignan ( )-SDG. LuPLR1 gene promoter consists of several putative regulatory boxes including the Myb1 box, a putative binding site for the LuMYB61 protein. This study demonstrated increase in DIR24, PLR1 and UGT74S1 expressions as well as increase in (+)-SDG production when LuMYB61 is overexpressed. We have also demonstrated when LuMYB61transcription factor is treated with NO donors, SNP or GSNO, S-nitrosylation occurs. It is a specific post translation modification which occurs on Cys residue present in the R2R3 domain of LuMYB61 proteins and as a consequence it abolishes DNA binding capacity and therefore decreases expression of DIR24, PLR1 and UGT74S1, key enzymes in phenylpropanoid metabolism thus leading to a decrease in lignan production.

Abstract (french)

Au cours de ces travaux, le rôle d’un facteur de transcription de la famille des MYB a été étudié, LuMYB61. Ce facteur contient deux domaines conservés R2R3 et son rôle dans le contrôle de la biosynthèse des lignanes a été suggéré. La Pinorésinol lariciresinol réductase 1, codée par le gène LuPLR1, est responsable de la biosynthèse du ( )-SDG, lignane le plus abondant du lin (Linum usitatissimum L.). Le promoteur du gène LuPLR1 abrite un élément cis-régulateur potentiel de fixation de facteurs MYB, l’élément Myb1. Cette étude a mis en évidence une augmentation coordonnée de l’expression des gènes DIR24, PLR1 et UGT74S1 ainsi que l'augmentation de la production de (+)-SDG en réponse à la surexpression de LuMYB61. Nous avons également démontré que la S-nitrosylation, une modification post-traductionnelle spécifique d’un résidu Cys présent dans le domaine R2 de ce facteur de transcription LuMYB61, induite par des donneurs de NO (le SNP ou GSNO) abolit sa capacité de liaison à l'ADN et par conséquent diminue l'expression des gènes DIR24, PLR1 et UGT74S1, des enzymes clés dans le métabolisme des phénylpropanoïdes conduisant ainsi à une diminution dans la production de lignanes.
