In vitro funkcionalno djelovanje bakterije Lactobacillus plantarum 16.4 na stanice HEp2

Abstract

U današnje vrijeme intenzivno se istraţuje primjena bakterija mliječne kiseline u prevenciji i liječenju raka. Mehanizmi koji bakterijama mliječne kiseline to i omogućuju su: utjecaj na fizikalno-kemijske uvijete i metaboličke aktivnosti u probavnom sustavu, vezanje i degradacija potencijalnih kancerogena, proizvodnja antitumorskih i antimutagenih spojeva te poboljšanje imunološkog odgovora domaćina. S ciljem dobivanja boljeg uvida u djelovanje bakterija mliječne kiseline na humane kancerogene stanice, testirano je djelovanje bakterije Lactobacillus plantarum 16.4 na stanice HEp2 te utjecaj L. plantarum na adhezijsko djelovanje koje enterobakterije Escherichia coli i Salmonella enterica pokazuju prema stanicama HEp2. Rezultati razlikovnog proteomskog istraţivanja pokazuju da u slučaju tretiranja stanica HEp2 s L plantarum nisu detektirani proteini tioredoksin, eIF5A te GSTP koji pospješuju rast kancerogenih stanica i njihovu otpornost na kemoterapeutike.

Abstract (english)

Nowadays a huge effort has been made in application and understanding of lactic acid bacterias (LAB) with anticancer activity. Mechanisms by which LAB may inhibit cancer are: influencing the physico-chemical conditions and the metabolic conditions in gastrointestinal tract of the host, binding and degradation of potential carcinogens, production of antitumor and antimutagenic compounds and enhancing the host's immune response. In order to get a better insight in LAB activity to human epidermoid larynx cancer cells (HEp2), the activity of Lactobacillus plantarum 16.4 to HEp2 cell line has been tested and the alteration of L. plantarum to adhesion properties of enterobacteria E. coli and S. enterica towards HEp2 cells. The results obtained from distinctive proteomics showed that in case of treating HEp2 cells with L. plantarum there is no expression of tioredoxin protein, eIF5A protein and GSTP who all participate in carcinogenesis and resistance to chemotherapeutics.
