Abstract

U ovom radu ispitivan je utjecaj ekspresije gena CCW12 iz Saccharomyces cerevisiae u kvascu Dekkera bruxellensis na svojstva njegove stanične stijenke. Ccw12p je manoprotein stanične stijenke kvasca S. cerevisiae koji pripada skupini proteina kovalentno vezanih u stijenku preko ostatka glikozilfosfatidilinozitol (GPI) sidra. U ovom radu dokazan je pozitivan utjecaj ekspresije CCW12 na brzinu rasta stanica, te je uočeno da stanice transformanata rastu u nakupinama. Unos gena CCW12 je djelovao pozitivno na sposobnost preživljenja stanica Dekkera bruxellensis u vodenom mediju, ali ne djeluje značajnije na osjetljivost stanica na 1M sorbitol, pa je u logaritamskoj fazi rasta čak i blago povećava. Transformirane stanice Dekkere bruxellensis su otpornije na djelovanje zimolijaze. Ovakavi rezultati govori u prilog da je unos gena CCW12 iz kvasca S. cerevisiae u stijenkama Dekkere bruxellensis vjerojatno izazvao promjene u strukturi stanične stijenke transformiranog soja.

Abstract (english)

This research is focused on effect of expression of CCW12 from Saccharomyces cerevisiae on the cell wall characteristics of Dekkera bruxellensis. Ccw12p is cell wall mannoprotein that belongs to the group of glycosylphosphatidylinositol anchor (GPI) associated proteins. In this work the positive effect of CCW12 expression on growth rate of Dekkera bruxellensis is proved. Transformed cells grow in clumps, and showed increased viability in water, but not in 1M sorbitol solution. Transformed cells of Dekkera bruxellensis are less sensitive to zymoliase. Such results indicate that transformation of Dekkera bruxellensis with CCW12 probably affected cell wall structure.
