Učinak ekstrakta biljke Amorpha fruticosa L. na humanim tumorskim staničnim linijama

Abstract

Proliferacija i adhezija životinjskih stanica ovisi o različitim faktorima rasta, hormonima i specifičnim proteinima koji su najčešće osigurani dodatkom seruma životinjskog podrijetla u medij za uzgoj stanica. Budući da primjena seruma ima brojne nedostatke, danas se sve češće nastoji smanjiti ili u potpunosti izbjeći njegova uporaba. Amorpha fruticosa L. je invazivna biljna vrsta prisutna u Hrvatskoj od početka 20. stoljeća čiji je biološki potencijal prepoznat te se sve više istražuje. In vitro ispitivanje učinka etanolnog ekstrakta biljke Amorpha fruticosa L. provedeno je na dvjema humanim tumorskim staničnim linijama (HeLa i MCF-7) te je određena vijabilnost stanica WST-1 metodom nakon 72 sata od tretmana. Ekstrakt amorfe pri ispitanim koncentracijama u rasponu 0,5-10 mg/mL nije imao citotoksično djelovanje. Pri najvećoj ispitanoj koncentraciji od 10 mg/mL zabilježen je pozitivan učinak na rast stanica, koji je nešto izraženiji kod MCF-7 nego kod HeLa stanica. Kada su stanice uzgajane u mediju s manjim volumnim udjelom seruma, 5 % naspram uobičajenih 10 % FBS, taj stimulacijski učinak je bio još izraženiji što ukazuje na potencijal primjene ove biljke kao alternative za zamjenu dijela seruma u mediju za uzgoj.

Abstract (english)

Proliferation and adhesion of animal cells is dependent on different growth factors, hormones and specific proteins, which are usually added to cultivation media by serum of animal origin. Since serum supplementation has many disadvantages, scientists try to reduce or completely omit its use nowadays. Amorpha fruticosa L. is an invasive plant species, present in Croatia since the beginning of 20th century, whose biological potential has been recognized and studied in recent years. In vitro study of the effect of ethanol extract of Amorpha fruticosa L. was conducted on human tumor cell lines (HeLa and MCF-7) and cell viability was determined by WST-1 method after 72 hours of treatment. Extract of Amorpha fruticosa L. was not cytotoxic in all tested concentrations ( 0.5-10 mg/ml). Stimulatory effect on cell growth was observed at the highest tested concentration of 10 mg/ml and it was slightly more pronounced in MCF-7 than in HeLa cells. When cells were grown in a medium with a reduced serum volume ratio, 5% vs.10% FBS, the stimulatory effect was even more prominent, indicating the potential use of this plant as an alternative for replacement of serum in culture medium.
