Abstract

Stanična stijenka kvasca Saccharomyces cerevisiae je ekstracelularna organela građena od polisaharida glukana i hitina te više od trideset različitih manoproteina. Scw4p se u staničnu stijenku veže na dva načina, tj. može se detektirati u frakciji kovalentno i u frakciji nekovalentno vezanih proteina stijenke. Također je poznatno da usljed procesiranja dolazi do različitog načina vezanja ovog proteina, pri čemu se neprocesirani protein veže kovalentnim vezama dok se procesirana forma veže nekovalentno. U ovom radu je dokazano kako Kex2p proteaza ne sudjeluje direktno u procesiranju Scw4p proteina, budući da do procesiranja ovog proteina dolazi i u stanicama kex2 mutanta, već da vjerojatno sudjeluje u aktivaciji japsinskih proteaza koje zatim procesiraju Scw4p. Također je potvrđeno kako na efikasnost procesiranja nisu utjecale mutacije uvedene u regiju SCW4 gena za koju se prema literaturnim podacima pretpostavljalo da je ciljna sekvenca za procesiranje Scw4p.

Abstract (english)

The cell wall of the yeast Saccharomyces cerevisiae is the extracellular organelle made up of polysaccharides glucans and chitin, and more than thirty different mannoproteins. Scw4p binds to the cell wall in two ways, so it can be detected in the fraction of covalently linked protein and in the fraction of nonconvalently linked cell wall protein. Different binding of this protein is consequence of processing, whereby the unprocessed protein binds covalently while processed form binds nonconvalently. Results in this paper showed that Kex2p protease does not participate directly in the processing of Scw4p protein, since the Scw4p is normaly processed in kex2 mutant cells, but is probably involved in the activation of yapsin proteases which then process Scw4p. Mutations introduced into the region of SCW4 gene, that according to literature is assumed to be the target sequence for Scw4p processing, did not affect processing efficiency.
