Abstract

ELISA testom i metodom tekućinskom kromatografijom visoke djelotvornosti u kombinaciji sa spektrometrijom masa (LC-MS/MS) analiziran je uzorak mlijeka obogaćen aflatoksinom M1 i na temelju rezultata uspoređene te dvije analitičke metode. Određen je parametar iskorištenja, koeficijent varijacije, linearnost metode i matriksa te limit detekcije određivanja aflatoksina M1. Rezultati su pokazali veću točnost analize LC-MS/MS metodom. Zbog toga se ELISA test i koristi kao orijentacijska metoda, dok LC-MS/MS služi kao potvrdna metoda u slučaju pozitivnog testa uzorka.

Abstract (english)

The sample of milk enriched with aflatoksin M1 has been analyzed by ELISA test and high performance liquid chromatography with tandem mass spectrometry detection (LC-MS/MS) and the comparation of these two analytical methods has been carried out based on the results. Recovery, coefficient of variation, linearity of method and matrix and detection limit of aflatoksin M1 detection have been determined. The results have shown higher measurement accuracy by LC-MS/MS analysis. Thus, ELISA test is used as a comparation method and LC-MS/MS as an affirmative method when the result shows to be positive.
