Abstract

U ovom istraživanju proučavan je postupak kemijske sterilizacije Hutnerove podloge za heterotrofni uzgoj alge E. gracilis. Vodikov peroksid i antimicin odobreni su kao sredstva za kemijsku sterilizaciju, a kvasac Saccharomyces cerevisiae i bakterija Bacillus subtilis kao predstavnici kontaminanata heterotrofnog uzgoja E. gracilis. Na osnovi rezultata istraživanja dobiveno je da je koncentracija vodikovog peroksida u intervalu od 4,5-7,5 mg/L optimalna za heterotrofni uzgoj E. gracilis pri čemu je uočeno usporavanje rasta kvasca i bakterije. Koncentracija antimicina od 0,2 g/L usporava rast E. gracilis i kontaminanata osobito bakterije B. subtilis a koncentracija od 0,5 g/L u potpunosti zaustavlja heterotrofni uzgoj E. gracilis. Na osnovi rezultata ovog istraživanja vidljivo je da je nužno daljnje istraživanje s ciljem definiranja optimalne koncentracije antimicina za heterotrofni uzgoj E. gracilis.

Abstract (english)

In this research chemical sterilization of Hutner medium for heterotrophic cultivation of E. gracilis was studied. Hydrogen peroxide and antimycin were selected as active compounds for chemical sterilization and yeast Saccharomyces cerevisiae and bacterium Bacillus subtilis as representitive contaminants of E. gracilis cultivation in heterotrophic cultivation. Respecticely on the basis of results obtained in this research it is evident that optimal H2O2 concentration interval for heterotrophic cultivation of E. gracilis is 4,5-7,5 mg/L where notable slow down of yeast and bacterium growth was observed. Antimycin concentracion of 0,2 g/L considerably slow down the growth of E. gracilis and contaminants (especially B. subtilis). However, antimycin concentracion of 0,5 mg /L completely stopped heterotrophic cultivation of E. gracilis. On the basis of this research it is clear that further investigation has to be performed in order to define the optimal antimycin concentracion for heterotrophic cultivation of E. gracilis.
