Abstract

U ovom radu ispitan je učinak planarnog PCB-77 i neplanarnog PCB-153 na proliferaciju CHO (Chinese Hamster Ovary)-K1 stanica metodom Neutral Red tijekom 72 sata. Za tretman su korištene različite koncentracije polikloriranih bifenila (10-100 µM). Pri nižim koncentracijama PCB-77 od 10 µM tijekom čitavog perioda inkubacije te 25 µM nakon 72 sata nije utvrđen statistički značajan inhibicijski učinak na proliferaciju stanica. PCB-153 inhibirao je staničnu proliferaciju tijekom cijelog perioda tretmana pri koncentracijama od 50100 µM. Prema izračunatim i uspoređenim vrijednostima za IC50, PCB-77 je imao izraženije citotoksično djelovanje od PCB-153. Ispitivanje mogućeg zaštitnog djelovanja antioksidansa vitamina E na vijabilnost stanica tretiranih sa 75 µM PCB-153 pokazalo je njegov protektivni učinak pri svim ispitanim koncentracijama (50, 75, 100 i 150 µM), međutim taj učinak nije bio statistički značajan, dok takvo protektivno djelovanje antioksidansa nije primijećeno pri tretmanu stanica s planarnim PCB-77.

Abstract (english)

In this study the cytotoxic effects of planar PCB-77 and nonplanar PCB-153 were examined by monitoring CHO (Chinese Hamster Ovary)-K1 cell proliferation using Neutral Red method during 72 hours. Different concentrations (10-100 µM) of polychlorinated biphenyls were used in treatment of cells. Statistically significant decrease of cell proliferation was not detected at lower concentrations of PCB-77 (10 µM) during whole period of incubation and of 25 µM after 72 hours. Higher concentrations (50-100 µM) of PCB-153 caused statistically significant decrease of cell proliferation during whole period of incubation. According to calculated IC50 values, PCB-77 was more cytotoxic than PCB-153. The study of possible protective impact of antioxidant vitamin E on the viability of cells treated with 75 µM PCB-153 showed that vitamin E has protective effect at all concentrations tested (50, 75, 100 and 150 µM), but the extent of effect was not statistically significant, while such protective effect of antioxidant is not observed when treating cells with planar PCB-77.
