Abstract

Rak dojke je prvi uzrok smrtnosti od raka kod žena. Razni genetski i okolišni čimbenici smatraju se osnovnim uzrocima raka dojke, što je genetski heterogena bolest. Rak nastaje zbog gubitka kontrole normalnog rasta stanica. Skupina gena upletenih u razvoj karcinoma su tumor supresorski geni, koji upućuju stanice da proizvode proteine koji kontroliraju rast i dijeljenje stanica. Ukoliko dođe do gubitka ili mutacije jednog od tumorsupresorskih gena tada može doći do nastanka tumora. U ovom radu istraživana je mikrosatelitna nestabilnost (MSI) i gubitak heterozigotnosti (LOH) tumor supresorskih gena APC, BRCA2 i MSH2 u tkivu pacijentica oboljelih od karcinoma dojke. Korištene su metode lančane reakcije polimeraze (PCR) i restrikcijskog cijepanja (RFLP). Istraživanjem uzoraka nije utvrđen MSI, dok je LOH dokazan u dva od 16 informativnih uzoraka gena APC i kod jednog od 23 informativnih uzoraka gena BRCA2.

Abstract (english)

Breast cancer is the first leading cause of cancer mortality in women. Various genetic and environmental factors have been established as causes of breast cancer, which is a genetically heterogeneous disease. Cancer arises from a loss of normal growth control. A group of genes implicated in cancer are the "tumor suppressor genes", genes that instruct cells to produce proteins that restrain cell growth and division. If one of tumor suppressor gene is mutated or lost, then the cancerous state may occur. In the present study, microsatellite instability (MSI) and loss of heterozygosity (LOH) in tumor supressor genes APC, BRCA2 and MSH2 were investigated in cancer patient tissues, using polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) methods. There were no case of MSI found, whereas two LOHs out of 16 informative samples of APC gene and one LOH out of 23 informative samples of BRCA2 gene were detected.
