Abstract

Klenbuterol je simpatotimetički amin i derivat katekolamina epinefrina i norepinefrina te pripada skupini lijekova zvanih β-agonisti. Uporaba β-agonista u anaboličke svrhe rezultira kumulacijom farmakološki aktivnih ostataka u mesu tretiranih životinja te su zabilježeni brojni slučajevi akutnih otrovanja kod ljudi nakon konzumacije jetre tretiranih životinja. U ovom radu istraživan je citotoksični i prooksidativni učinak klenbuterola na humanim stanicama u kulturi, genotoksični učinak na ljudske limfocite in vitro, te genotoksični učinak na stanice jetre i krvi zamorčadi in vivo. Istraživanje citotoksičnosti i prooksidativnog učinka na humanim stanicama u kulturi pokazalo je potencijalno mitogeni učinak pri dozi 10 puta većoj od NOAEL vrijednosti, te prooksidativni učinak pri istoj koncentraciji. Pri koncentraciji od 25 μg/L klenbuterol izaziva lomove u molekuli DNA ljudskih limfocita tretiranih u uvjetima in vitro. U in vivo uvjetima, klenbuterol je bio izrazito citotoksičan na stanice jetre zamorčadi, dok se u preživjelim stanicama nije pokazao njegov genotoksični karakter. U limfocitima zamorčadi klenbuterol se pokazao kao genotoksičan spoj u ovisnosti o koncentraciji.

Abstract (english)

Clenbuterol is a symphatomimetic amine derivative of catecholamines epinephrine and norepinephrine and belongs to a class of drugs called β-agonists. The use of β-agonists in anabolic purposes resulted in acumulation of pharmacologically active residues in meat of treated animals, and there were many recorded cases of acute poisoning in humans after consumption of the liver of treated animals. In this thesis, we investigated the cytotoxic and prooxidant effects of clenbuterol in human cells in culture, genotoxic effects on human lymphocytes in vitro, and evaluated the effects on the cells of the liver and blood of guinea pigs in vivo. Research of cytotoxic and prooxidative effect on human cells in culture showed potentially mitogenic effect at a dose 10 times higher than the NOAEL values and prooxidant effect at the same concentration. At a concentration of 25 μg/L, there were DNA aberations in human lymphocytes in vitro. In vivo, clenbuterol was extremely cytotoxic to liver cells of guinea pigs, while in the surviving cellsno genotoxicity was observed . In lymphocytes of guinea pigs clenbuterol has proven to be a genotoxic compound dependent on the concentration applied.
