Abstract

Adhezija probiotičkih sojeva na epitelne stanice gastrointestinalnog trakta (GIT) jedna je od najvažnijih karakteristika probiotičkih sojeva jer omogućuje kolonizaciju GIT domaćina, gdje se očekuju pozitivni učinci na zdravlje domaćina. Probiotički soj Lactobacillus helveticus M92 posjeduje površinski S-layer protein, koji je identificiran pomoću SDS-PAGE u kombinaciji s masenom spektrometrijom, a za koji se smatra da je jedan od čimbenika koji sudjeluju u adheziji bakterija na epitelne stanice domaćina. Stoga je u ovom radu ispitana adhezija L. helveticus M92 na Caco-2 epitelne stanice, prije i nakon uklanjanja S-layer proteina s površine bakterijske stanice pomoću litijevog klorida. Uspoređivanjem broja adheziranih probiotičkih stanica, koji je nakon uklanjanja S-layer proteina bio značajno smanjen, utvrđena je moguća uloga ovog površinskog proteina na adheziju L. helveticus M92 na epitelne Caco-2 stanice.

Abstract (english)

Adhesion of the probiotic strains on the epithelial cells of the gastrointestinal tract (GIT) is one of the most important characteristic of probiotic strains because it allows colonization of the host GIT, where are expected postive effects on the host's health. Probiotic strain of Lactobacillus helveticus M92 has a surface S-layer protein which was identified by SDS-PAGE combine with mass spectrometry, and which is considered to be one of the factors involved in adhesion of bacteria to host epithelial cells. Therefore, in this study we had examined adhesion of L. helveticus M92 on Caco-2 epithelial cells before and after removal of the S-layer proteins from the surface of bacterial cells using lithium chloride. Comparing the number of the adhered probiotic cells, which after removal of S-layer protein was significantly reduced, was determined the possible role of this protein on the surface adhesion L.helveticus M92 on epithelial Caco-2 cells.
