Factors influencing the success of microspore and anther culture for wheat and triticale haploid production

Abstract

Anther culture technique is employed in laboratory 'Le Menhir' for production of triticale and wheat haploid plants. Microspore culture technique shows a potential to be better than anther culture in haploid production; therefore, it is evaluated in laboratory 'Le Menhir'. This study is one of the steps towards the development of successful microspore culture protocol as well as the part of an evaluation process. In this work, no haploid plants were produced using microspore culture protocol; although microspores from more than 400 spikes were put in culture. This is probably due to the bad health state of donor plants and genetic effects. The haploids were produced using anther culture protocol. The average production of triticale haploids is 1.82 green plants per spike and wheat haploid 0.33 green plants per spike. Genotypic effects on production success are obvious and they are presented in the work. Furthermore, the parent genotypes are classified by the average success rates of their progeny, which genotype’s 15580 progeny has the best average success rate. This data will be used in future experiments. In addition, the development of microspores towards embryos is described. As the next step verification of these results is proposed as well as modifications of the microspore culture protocol.
